The principle of fluorescence microscopy is based on the property of luminescence. Basically, luminescence refers to the ability of material to emit radiation of its own after being subjected to an exciting light (ultra-violet, blue, or green). One type of luminescence is called phosphorescence. Materials with this property have the ability to maintain some radiation emission after cessation of the exciting radiation. The other type of luminescence is what drives the fluorescence microscope and is known as fluorescence. When this type of material has the exciting radiation stopped, the substance also stops radiating.
Wavelength charts are important to understanding the principles and practices used in fluorescence microscopy. Emitted radiation from the specimen is longer than the excitation radiation. The exciting wavelength is of a short wavelength such as from ultra-violet or blue light. The molecules in the specimen absorb the exciting radiation, then release emitted radiation, which is then passed through filters and on to the eyepieces in the fluorescence microscope. Fluor chromes are a type of substance that is able to emit radiation when exciting radiation is incident upon it.
Fluorescence microscopes utilize a high intensity light source such as in lamps made of tungsten-halogen, quartz-iodine, and high pressure mercury. These types of lamps produce in the 300-350nm range and includes the shorter wavelength ultra-violet light.

The excitation filter in the dichroic filter cube of the fluorescent microscope should cut out all but a narrow bandwidth of wavelengths that match the maximum excitation range of the fluorochrome. The emission filter narrows the emitting wavelengths to a small band is also custom designed for the fluorescence application.
